Özet
Diazo bileşikleri, organik bileşiklerin ilginç bir grubunu oluştururlar ve karben ve karbenoidlerin başlıca kaynaklarındandırlar. ?-Diazo-?-okso-alde-hitler, bazı doğal bileşiklerin, antibiyotiklerin ve triazol türevlerinin sentezinde kullanıla-bilmektedir. Bununla birlikte 1994 yılına kadar bu bileşiklerin yalnızca üç örneği bilinmekteydi. Sezer ve Anaç 1994 yılında bir azidopiridinyum tuzu kullanarak orijinal bir diazo transfer reaksiyonu ile, ve daha sonraki bir çalışmalarında diazometil ketonların Vilsmeier–Haack formilasyonu ile bu bileşikleri çoğaltılmışlardır. Her iki yöntemin dezavantajlarından dolayı açildiazoasetaldehitlerin sentezi için alternatif bir yönteme gereksinim duyulmuştur: Bu çalışmada, iki farklı azidoformamidinyum tuzu hazırlanmış ve bu tuzların benzoilasetaldehitle nötral-asidik ortamda reakiyonu ile açil-diazoasetaldehit sentezi amaçlanmıştır. Tetrametilazidoformamidinyum klo-rü-rün benzoilasetaldehit ile reaksiyonu sonucunda herhangi bir yan ürün oluş-turmaksızın benzoildiazoasetaldehit tek cins diazo bileşik olarak elde edilmiştir. Benzer şekilde 4-izopropoksibenzoilasetaldehit’in reaksiyonundan yine tek ürün olarak 4-izopropoksibenzoildiazoasetaldehit elde edilmiştir. Bu sonuçlar, tetra-metilazidoformamidinyum klorürün ?-diazo-?-oksoasetaldehitlerin sentezinde kullanı-labilecek uygun bir diazo transfer reaktifi olduğu konusunda ümit vericidir.
Diazo compounds constitute an interesting class of organic compounds and are the main precucors of carbenes of carbenoids. Acyldiazoacetaldehydes, which can be used to synthesize some natural compounds, antibiotics and triazole derivates, constitute one class of diazo compounds. Only three examples of acyldiazoacetaldehydes were known until 1994 at which time Sezer and Anaç synthesized many of them employing a novel diazo transfer reaction using an azidopyridinium salt and Vilsmeier-Haack formylation of diazomethyl ketones. Because of the disadvantages of the both methods, an alternative method is necessary for the synthesis of ?-diazo-ß-oxoaldehydes. In this study, we prepared two different azidoformamidinium salts and tried the reactions of these salts with benzoyl-acetaldehydes in neutral-to-acidic medium in order to synthesize ?-diazo-ß-oxoaldehydes. The reaction between tetramethyl-azido-formamidinium chloride and benzoylacetaldehyde gave only one diazo compound, that is benzoyldiazoacetaldehyde, without any detectable presence of by-products. Similarly, the reaction of 4-isopropoxybenzoylacetaldehyde with tetramethyl-azido-formamidinium chloride gave 4-isopropoxybenzoyl-diazo-acet-aldehyde as the only diazo product. These results are very promising that tetra-methyl-azido-formamidinium chloride can be used as a diazo transfer reagent for the direct syntheses of ?-diazo-?-oxoaldehydes.
