Özet
Bu çalışmanın amacında insan sağlığı ve gıda maddeleri için önemli olan doğal antioksidanlardan flavonoidlerin analizi için basit, tekrarlanabilirliği yüksek, reaksiyon basamakları az, ucuz ve spesifik olmayan cihazlarla yapılan bir yöntem geliştirilmek istenmiştir. Flavonoidler antioksidan aktivite gibi biyolojik bakımdan önemli özellikler gösterirler. Doğal antioksidanlardan flavonoidler vücudumuz tarafından üretilemediklerinden, sebze ve meyveleri tüketerek bünyemize almaktayız. Antioksidanlar, canlı metabolizmalarında bulunan serbest radikaller ve reaktif oksijen türleri ile arasındaki dengenin bozulması sonucu oluşan oksidatif stresin yaratmış olduğu zararı azaltıcı rol oynayan moleküllerdir. Metot sülfat asitli ortamda Ce(IV)’ün maksimum dalga boyundaki (320 nm) absorbans değeri, Ce(IV)’ün flavonoidler ile reaksiyonu sonucu azalmaktadır. Ce(IV)’ün maksimum dalga boyundaki bu azalmadan yararlanarak indirekt olarak toplam flavonoid miktarını hesaplayabilmektedir. Bu çalışmanın ikinci kısmında ise, flavonoidlerin, Ce(IV) ile reaksiyonu sonucu oluşan Ce(III)’ün emisyon değerlerinin ölçülmesine dayanan spektroflorotometrik bir yöntem kullanıldı. Ce(IV) floresans özelliğe sahip değil iken Ce(III) floresanas özelliğe sahiptir. Böylece Ce(IV)’ün flavonoidler ile reaksiyonu sonucu oluşan Ce(III)’ün emisyon değerleri okunarak direkt olarak toplam flavonoid miktar tayini yapıldı. Isırgan otuna ve çeşitli bitki ekstraktlarına analiz yöntemlerinde belirtilen prosedür başarıyla uygulandı. Sonuçları literatürdeki çalışmayla karşılaştırdığımızda, uyumlu sonuçlar elde edilmiştir.
The aim of this study is to the develop a method which is simple for analyzing flavonoids from natural antioxidants that are important for human health and nutrients and with high repeatable with less number of reaction, applicable by cheap and non-specific equipments. Flavonoids have been shown to have a number of biologically important roles such as antioxidant activity. Flavonoids cannot be produced by the human body and thus have to be supplemented through the daily diet. Antioxidants may serve the task of reducing oxidative damage in humans induced by free radicals and reactive oxygen species under ‘oxidative stress’ conditions. The method is based on the maximum absorbance value of Ce (IV) at the wave length of 320 nm in acidic medium decreases during the reaction of Ce (IV) with flavonoids. Thus, the total amount of flavonoid can be calculated indirectly depending on the decreased absorbance value. In the second part of this study, spectroflourometric method which is based on measuring the emission of Ce(III) that appears as a result of the reduction reaction between flavonoids and Ce(IV) ions which is non-fluorescent. Thus, total flavonoid amount was possible to calculate by measuring the fluorescence intensity of the Ce(III). The procedure was successfully applied to the assay of total flavonoids in the aerial parts of Urtica Dioica L. and the extract of different plants. The comparison of the results with the official method given in the literature was in a good agreement.
