Özet
Polimerik nanopartiküller, uzun dolaşım süreleri ve terapötik ajanların taşınmasına olanak sunmaları sayesinde, son zamanlarda özellikle kontrollü ilaç salım sistemleri ve gen taşıma alanlarında büyük ilgi görmektedir. Taşıyıcı sistemler için yapılan araştırmalarda özellikle kanser dokularına hedeflenme yaklaşımı ile akıllı ve nanoboyutlu taşıyıcıların geliştirilmesine odaklanılmıştır. Polimerik nanopartiküllerin özel olarak tasarımı, çoğunlukla istenen uyarıcıya yanıt veren yapıların polimerlere dahil edilmesiyle olur. Bununla birlikte, polimerlerin iyi tanımlanmış ve yeniden üretilebilir bir şekilde sentezlenmesi polimerik nanopartiküllerin tasarımı için önemlidir. Son yıllarda, polimerik salım sistemleri arasında, Poli(2-oksazolin)'ler (genellikle PAOx, POZ, POx veya POXA olarak kısaltılır), benzersiz yapısal özelliklerinden dolayı büyük ilgi görmüştür. Poli (2-oksazolin)'ler, çok yönlü ayarlanabilir özelliklere ve mükemmel biyouyumluluğa sahip olağanüstü polimer platformunlarını temsil eder ve bu da onları çok çeşitli uygulamalarda kullanılabilir hale getirir. Nanopartiküller, genellikle lipozom, misel, polimerzom, nanojel ve polimer-ilaç konjugatlarını temsil eder. Poli(2-oksazolin) (POx) esaslı polimerik terapötiklerin geliştirilmesi, yeni nesil polimerik nanopartikül platformların hazırlanmasına olanak sağlamıştır. Bu tez, ilaç/gen kapsülleme ve salım sistemleri için üç farklı yeni PEtOx esaslı, kansere hedefenebilir polimerik nanopartiküllerin tasarımını, sentezini, karakterizasyonunu ve optimizasyonunu sunmaktadır. Farklı yapıda polimerik nanopartiküllerin sentezi için üç farklı sentetik strateji geliştirilmiştir. İlk çalışmada, DOPE lipid yapısına PEtOx polimeri entegre edilerek kanser hücrelerine özgül olarak hedeflenebilen peptit 18 ve peptit 563 ile modifiye edilen lipopolimerlerin hazırlanabilirliği ve böylece gen transferi için potansiyel kansere hedeflenebilir taşıyıcı sistemin geliştirilebildiği gösterildi. İkinci diğer çalışmada, misel formülasyonları için P(EtOx-ko-EI)-b-PCL blok kopolimerler hazırlandı. Bu çalışmada hidrofilik blok olarak P(EtOx-ko-EI) ve hidrofobik yapı için PCL bloğu içeren yeni bir amfifilik ve katyonik özellikte blok kopolimeri tasarlandı ve klik reaksiyonları kullanılarak peptitlerle konjuge edildi ve sonra polimerik miseller hazırlandı. Üçüncü ve son çalışmada ise çok işlevli hale getirmeye yatkın PEtOx tabanlı nanojellerin tasarımı ve sentezi yapıldı. PEtOx üç kollu başlatıcı kullanılarak yaşayan katyonik halka açma polimerizasyonu ile yıldız şeklinde hazırlandı. Daha sonra azid-alkin konjugasyonu ile alkin içeren flüoresan etiket (BODIPY) yıldız PEtOx polimere konjuge edildi. Nanojelleri sentezlemek için ditiol bazlı bir çapraz bağlayıcı kullanıldı. Jelleşme sonrası azid fonksiyon grupları içeren nanojel hedefleme ajanı peptit 563 ile konjuge edildi. İlaç salımı elde etmek için nanojeller, kemoterapötik ilaç adayı SGK 636 ile yüklendi ve ilaç yükleme kapasitesi incelendi.PEtOx polimerler kolay bir şekilde hazırlanarak ve etkili konjugasyon kimyası kullanılarak çok işlevli hale getirilebilir ve terapötik ajanlar olarak yapılandırılabilir. Sonuç olarak, PEtOx'lara dayanan iyi tanımlanmış polimerik taşıyıcı sistemler, kanser tedavisi uygulamaları için umut verici adaylar olarak hizmet edebilir.
Polymeric nanoparticles have attracted great attention, especially in the fields of controlled drug delivery systems and gene transport due to their long circulation times and the ability to transport therapeutic agents. In researches for carrier systems, especially with the targeting approach to cancer tissues, it has been focused on the development of smart and nano-sized carriers. The special design of polymeric nanoparticles is often by incorporating structures that respond to the desired stimulus into polymers. However, polymers in a well-defined and reproducible manner are important for the design of polymeric nanoparticles. In recent years, among polymeric release systems, Poly(2-oxazoline)s (usually abbreviated as PAOx, POZ, POx or POXA) have attracted great attention due to their unique structural properties. Poly(2-oxazoline)s represent exceptional polymer platforms with versatile adjustable properties and excellent biocompatibility, making them usable in a wide variety of applications. Nanoparticles generally represent liposome, micelle, polymersome, nanogel, and polymer-drug conjugates. The development of poly(2-oxazoline)(POx) based polymeric therapeutics has enabled the development of a new generation of polymeric nanoparticle platforms. This thesis presents the design, synthesis, characterization and optimization of three different new PEtOx-based cancer-targetable polymeric nanoparticles for drug/gene encapsulation and delivery systems. Three different synthetic strategies have been developed for the synthesis of polymeric nanoparticles of different structures. In the first study, it was demonstrated that by integrating PEtOx polymer into the DOPE lipid structure, the preparation of peptide 18 and peptide 563 modified lipopolymers that can be specifically targeted to cancer cells can be developed, and thus the potential cancer targetable carrier system for gene transfer can be developed (Figure 1). Surface modifications of liposome enable variety advantages, such as, increasing stability, and targeted drug, gene, and image agent delivery. Peptide 18/peptide 563-poly(2-ethyl-2-oxazoline)-dioleoylphosphatidyl-ethanolamine (P18/ P563-PEtOx-DOPE) have been successfully prepared. P18/P563-PEtOx-DOPE were defined by FT-IR spectroscopy, GPC, and 1H-NMR. In this design, PEtOx conjugated liposomes with molecular weights of 3,500 Da and 5,800 Da were firstly synthesized. Better particle size (88.74 ± 0.6816) was obtained in PEtOx5800-DOPE according to DLS results. Then, peptide-18, peptide-563, and 7-mercapto-4-methyl coumarin were conjugated to the PEtOx5800-DOPE. The genetic material was encapsulated into liposomes. Migrated of genetic material was tracked by agarose gel electrophoresis. As a result, a gene transfer system with great potential was presented with this study. In the second other work, P(EtOx-co-EI)-b-PCL block copolymers were prepared for micelle formulations. In this study, a new amphiphilic and cationic block copolymer containing P(EtOx-co-EI) as a hydrophilic block and PCL block for the hydrophobic structure was designed and conjugated with peptides using click-type reactions, and then polymeric micelles were prepared (Figure 2). Here we prepared self-assembled polymeric micelles with targeted agents as potential carrier systems. The breast and prostate cancer specific targeting peptide modified poly(2-ethyl-2-oxazoline)-co-polyethyleneimine-block-poly(-caprolactone) block copolymers based micelles have been presented as a targetable carrier system. P(EtOx-co-EI) copolymers containing PEI as 30% and 60% hydrolysis degrees of PEtOx were prepared. P(EtOx-co-EI)-b-PCL copolymers were synthesized by copper(I)-catalyzed azide–alkyne cycloaddition click reaction. Then, peptide 18 and peptide 563 were modified to block copolymers via a thiol–ene click-type reaction. The prepared peptide conjugated block copolymers were confirmed by 1H-NMR, FT-IR, UV-Vis spectrometry and GPC. Micelles by self-assembly prepared with peptide conjugated and and non-peptide block copolymers with two different chain lengths of PEI blocks have given the particle sizes between 82 ± 0.6 and 170 ± 10.7 nm. The micelles having 60% PEI block demonstrated the highest positive zeta potential values.In the third, the design and synthesis of PEtOx-based nanogels that are prone to multi-functionalization were carried out (Figure 3). PEtOx was prepared in a star shape by living cationic ring-opening polymerization using a three-arm initiator. Then, the fluorescent label containing alkyne (BODIPY) was conjugated to star PEtOx polymer by azide-alkyne conjugation. A dithiol based crosslinker was used to synthesize the nanogels. After gelation, the nanogel targeting agent containing extra azide functional groups was conjugated with peptide 563. Subsequently, the nanogels were loaded with the chemotherapeutic drug candidate SGK636 and the drug loading capacity was examined. The size of the nanogel could be controlled at 100-200 nm with relatively narrow size distributions. The nanogels with size of 142.6± 19 nm were verified by proton nuclear magnetic resonance (NMR), Fourier transform infrared spectroscopy (FT-IR), dynamic laser scattering (DLS) and environmental scanning electron microscopy (ESEM). SGK 636, drug for prostate cancer treatment, was loaded into nanogel with high drug loading efficiency up to %68,7. In vitro drug release studies showed that up to 30,87% SGK 636 from nanogel was released in 24 h because of breaking disulfide bonds and swelling of nanogel. In addition, physical stability nanogel formulations in the physiological environment has been examined at pH 7.4 and %1-5 fetal bovine serum.PEtOx polymers can be easily prepared and multifunctionalized using conjugation chemistry and therapeutic agents. As a result, well-defined polymeric carrier systems based on PEtOx's can serve as promising candidates for cancer therapy applications.
