Özet
Çalışmada pirinç kepeği, buğday kepeği, ayçiçeği tohumu ve ayçiçeği tohumu küspesinden izole edilen küfler ile bir merkezden temin edilen küf kültürü kullanılmıştır. Bu substratlar üzerinde katı faz fermentasyon tekniği ile geliştirilen küflerin lipaz aktiviteleri saptanmıştır. Küfler arasında en yüksek lipaz aktivitesini (462,5±25,96 U) ayçiçeği tohumundan izole edilen Rhizopus spp. cinsi küfün gösterdiği gözlenmiştir. Lipaz aktivitesinin en yüksek olduğu substratlar ayçiçeği tohumu küspesi ve pirinç kepeği olarak belirlenmiştir. Çalışmada tripton, amonyum klorür ve maya ekstraktı gibi farklı azot kaynaklarının küf gelişimi ve lipaz aktiviteleri üzerindeki etkileri de ayrıca incelenmiştir. En yüksek enzim aktivitesine (580±3,53 U) pirinç kepeğinin substrat, amonyum klorürün azot kaynağı olarak kullanıldığı koşullarda 30°C’de 120 saatlik inkübasyon sonucunda ulaşılmıştır. Fermantasyon ortamına amonyum klorür (% 0,75) yanında zeytinyağı (%1) ve su (%60) ilave edilmiştir. Bu koşullarda üretilen Rhizopus spp.’den elde edilen lipaz enziminin pH 7 ve 37°C sıcaklıkta en yüksek spesifik aktiviteyi gösterdiği belirlenmiştir. Amonyum sülfat ile çöktürme yoluyla kısmi saflaştırılarak elde edilen lipaz çökeltisinin hidroliz kabiliyetinin ticari Lipozyme RM IM (Rhizomucor miehei) ile aynı, asidoliz reaksiyonundaki etkinliğinin ise referans lipaza göre yarı değerlerde olduğu gözlenmiştir.
The fungi isolated from wheat and rice bran, sunflower seed and sunflower seed wastes and one mould species obtained from a culture collection were used in this study. Moulds were previously screened for their lipase activities following their growth on these solid substrates. Among the moulds Rhizopus spp. isolated from sunflower seed exhibited the highest lipase activity (462,5±25,96 U). Sunflower seed waste and rice bran were found to be the most suitable substrates for lipase production. The effects of different nitrogen sources including tryptone, yeast extract and ammonium chloride on the growth and lipase production of the moulds were also investigated. The highest yields of enzyme activity (580±3,53 U) was obtained on rice bran substrate at the end of 120 h/30°C of incubation when ammonium chloride was used as the nitrogen source. The rice bran substrate contained olive oil (1%), water (60%) and ammonium chloride (0,75%). The optimum pH and temperature values for enzyme activity were found to be 7 and 37°C respectively. Rhizopus spp. lipase precipitate obtained by partially purifying with ammonium sulfate showed similar hydrolysing capacity to Lipozyme RM IM (Rhizomucor miehei) whereas activity in acidolysis reaction was half of the reference lipase enzyme.
