Farklı Ekstraksiyon Metotları İle Adaçayı  (salvia Officinalis L.) Bitkisinden Antioksidan Ekstraksiyonunun Optimizasyonu

Özet
Son yıllarda tüketicilerin doğal ürünlere olan ilgisinin artması ve yapay antioksidanların sağlık üzerine olumsuz etkileri nedeniyle kullanımlarının sınırlandırılması ile oksidatif bozulmayı engellemek üzere bitkisel kaynaklardan elde edilen doğal antioksidanların gıdalarda koruyucu olarak kullanılmasına yönelik araştırmalar artmıştır.  Serbest radikaller gıdalarda  neden oldukları bozunma reaksiyonlarının dışında, canlı hücre ve organ sistemleri üzerinde kanserojen, erken yaşlanma, kalp damar hastalıkları gibi önemli sağlık etkilere neden olmaktadır. Bu doğrultuda serbest radikallerin neden oldukları olumsuz sağlık etkilerini yavaşlatmak ya da durdurmak için antioksidan içeriği yüksek olan gıdaların tüketilmesi gerektiğinden bu ürünlere ilgi ve talep artmıştır. Antioksidan özelliklerinin yüksek olması nedeniyle Lamiaceae familyasına ait  biberiye, adaçayı, kekik, nane gibi aromatik bitkiler son yıllarda birçok araştırmaya konu olmuştur. Adaçayı (Salvia officinalis L.) çok eski çağlardan beri şifalı bir bitki olarak bilinmektedir. Adaçayının sağlık üzerine bilinen etkilerinden bazıları şu şekildedir; uyarıcı, iştah açıcı, antimutajen, ateş düşürücü, spazm çözücü, sindirimi kolaylaştırıcı, soğuk algınlığını iyileştirici, antimikrobiyal. Adaçayının temel antioksidan etkisi karsonik asit, karnosol, kafeik asit türleri ve rosmarinik asit bileşenlerinden gelmektedir. Bu çalışmada antioksidan özelliğinin yüksek olduğu bilinen adaçayından (Salvia officinalis L.) farklı ekstraksiyon metotları kullanılarak antioksidan ekstraksiyonu yapılmıştır. Kullanılan yöntemler; klasik çözücü, mikrodalga destekli, ultrason destekli ekstraksiyondur. Yanıt yüzey yöntemi kullanarak, her bir ekstraksiyon yöntemi için optimum ekstraksiyon koşulları belirlenmiştir. Bağımsız değişkenler; klasik çözücü ekstraksiyonunda; etanol konsantrasyonu (%60-80), sıcaklık (40-60 ˚C) ve ekstraksiyon süresi (60-120 dakika) olarak; ultrason destekli ekstraksiyonda;  etanol konsantrasyonu (%60-80), sıcaklık (40-60 ˚C) ve ekstraksiyon süresi (15-45 dakika) olarak; mikrodalga destekli ekstraksiyonda; etanol konsantrasyonu (%60-80), mikrodalga gücü (300-600 Watt) ve ekstraksiyon süresi (30-90 dakika) olarak seçilmiştir. Yanıt olarak toplam fenolik miktarı, toplam flavonoid miktarı ve DPPH (1,1-difenil-2- pikrilhidrazil) radikali yakalama yöntemi ile antioksidan aktivitesi seçilmiştir. Sabit tartıma gelene kalar kurutulan adaçayı bitkisi, belirlenen her bir ekstraksiyon yöntemi için farklı ekstraksiyon koşulları altında Box-Behnken deney tasarımına göre merkez noktada 3 tekrarla toplamda 15 deney noktasında ekstrakte edilmiştir. Elde edilen ekstraktlar 2500 rpm de 20 dakika santrifüj edilmiş, üst faz alınarak gerçekleştirilecek analizlere kadar -20˚C'de muhafaza edilmiştir. Toplam fenolik madde analizi Folin-Ciocalteau yöntemine göre yapılmış, sonuçlar gallik asit cinsinden verilmiştir. Toplam flavonoid madde analizi alüminyum klorür kolorimetrik metoduna göre gerçekleştirilmiş ve antioksidan kapasite miktarı, DPPH radikali yakalama yöntemine göre yapılarak sonuçlar toplam flavonoid madde için kateşin cinsinden, antioksidan kapasite için trolox cinsinden hesaplanmıştır. Klasik ekstraksiyon yönteminde, belirlenen optimum koşullarda ekstrakte edilen toplam fenolik madde, toplam flavonoid miktarı ve antioksidan kapasitesi sırasıyla, 3847.10 mg GAE/100 g kuru madde, 3250.90 mg CE/100 g kuru madde, 506.28 mg TEAC/100 g kuru madde olarak tespit edilmiştir. Klasik ekstraksiyon yöntemi ile adaçayından antioksidan ekstraksiyonu için belirlenen optimum koşullar; %60 etanol konsantrasyonu, 53.93  ˚C ekstraksiyon sıcaklığı, 84.84 dakika ekstraksiyon süresidir. Ultrason destekli ekstraksiyon yönteminde belirlenen optimum koşullarda ekstrakte edilen toplam fenolik madde, toplam flavonoid miktarı ve antioksidan kapasitesi sırasıyla, 4196.00 mg GAE/100 g kuru madde, 3706.00 mg CE/100 g kuru madde, 496.88 mg TEAC/100 g kuru madde olarak tespit edilmiştir. Ultrason destekli ekstraksiyon yöntemi ile adaçayından antioksidan ekstraksiyonu için belirlenen optimum koşullar; %60 etanol konsantrasyonu, 60  ˚C ekstraksiyon sıcaklığı, 38.03 dakika ekstraksiyon süresidir. Mikrodalga destekli ekstraksiyon yönteminde, belirlenen optimum koşullarda alınan sonuçlar ise toplam fenolik madde, toplam flavonoid miktarı ve antioksidan kapasitesi sırasıyla, 6271.00 mg GAE/100 g kuru madde, 5033.00 mg CE/100 g kuru madde, 493.22 mg TEAC/100 g kuru maddedir. Mikrodalga destekli ekstraksiyon yöntemi ile adaçayından antioksidan ekstraksiyonu için belirlenen optimum koşullar; %60 etanol konsantrasyonu, 518.18 Watt mikrodalga gücü, 82.72 saniye ekstraksiyon süresidir. Uygulanan üç yöntem karşılaştırıldığında, mikrodalga destekli ekstraksiyon yönteminin en verimli yöntem olduğu görülmektedir. Bunu ultrason destekli ekstraksiyon ve klasik çözücü ekstraksiyonu takip etmektedir. Bu çalışmadan elde edilen sonuçlar neticesinde, kısa süreli ekstraksiyonla yüksek antioksidan elde edilen mikrodalga destekli ekstraksiyon yönteminin adaçayından antioksidan ekstrakte etmek için kullanılabileceği söylenebilir.
Lipid oxidation is one of the major problem occurring in oils and foods rich in polyunsaturated fatty acids. In order to control lipid oxidation, synthetic compounds have been widely used in food industry. Nevertheless, toxicological effects together with consumer preference for natural products have resulted in increased interest in the use and research of natural antioxidants. Plant antioxidants protect human tissues against stress and diseases as well as they prevent the oxidation of foods. Presence of phenolics help protect against cardiovascular diseases, cancer and other degenerative diseases. The use and consumption of natural antioxidants from plant material has become an important to prevent from disease. Phenolics are complex molecules found in all plants as their secondary metabolites.  These include simple phenols, hydroxybenzoic acid and cinnamic acid derivatives, flavonoids, coumarines and tannins, among others. Phenolics can complex with carbohydrates, proteins, and other compunds which can influence the extraction effeciency. Phenolics are also not uniformly distributed in plants at the tissue, cellular and subcellular levels. Insoluble phenolics are the components of cell walls, while soluble phenolics are compartmentalized within the plant cell vacuoles. This is why all phenolics or phenolic specific groups is not possible to extract completely from plant material. Extraction parameters such as solvent selection and concentration, particul size of sample, extraction temperature, extraction time, solid:solvent ratio are effect the extraction yield of phenolic compounds. Extraction method is also important for extraction phenolic compounds from plant materials. Because of some drawback of solvent extraction, new extraction processes have been developed with the aim of providing characteristics such as enhanced selectivity, automation, lower consumption of organic solvents, and higher extraction efficiency. Techniques such as supercritical fluid extraction, pressurized liquid extraction, subcritical water extraction, ultrasound-assisted extraction, microwave-assisted extraction, solid-phase extraction, solid-supported liquid–liquid extraction, high hydrostatic pressure extraction, matrix solid-phase dispersion, and countercurrent chromatography are some advanced extraction techniques that have been applied to phenolic compound extraction, isolation, and fractionation. Response surface method (RSM) is a mathematical and statistical tool, which has been widely used to optimise various parameters in industry processing. RSM can evaluate the impact of different multiple parameters, and simultaneously optimise experimental conditions.  Box–Behnken design (BBD), a type of RSM, is a second-order multivariate technique based on three-level incomplete factorial design. BBD has been widely applied in the past decade to optimize the extraction procedure of bioactive compounds from natural sources, such as phenolic compounds. The interest has been increased to medicinal and aromatic plants due to high antioxidant activity. Sage (Salvia officinalis L.) is one of the most important medicinal and aromatic plant which belongs to Lamiacaea family. Recently, they have been extensively studied for their antioxidant activities as well. Sage is widely also used in traditional medicine since ancient times for their health beneficial effects such as stimulating, appetizing, antimutagenic, and fever reducer, anti-spasmodic, digestive, colds healing, antimicrobial. The main phenolic compounds identified in sage samples are rosmarinic acid, carnosic acid, carnosol, rosmanol. In this study, solvent extraction, ultrasound assisted extraction and microwave assisted extraction techniques were applied for the extraction of antioxidant compounds from sage (Salvia officinalis L.). Response surface methodology was employed to optimize the extraction conditions according to the total phenolic content, total flavonoid content and total antioxidant capacity (DPPH method). Ethanol concentration (60-80%), extraction  temperature (40-60 °C), extraction time (60-120 min) for solvent extraction; ethanol concentration (70-90%), extraction temperature (40-60 °C), extraction time (15-45 min) for ultrasound assisted extraction;  and ethanol concentration (70-90%), microwave power (300-600 Watt), extraction time (30-90 s) for microwave assisted extraction were selected as the independent variables. Total phenolic content, total flavonoid content and total antioxidant capacity were selected for response.  Fresh sage was dried in an oven at 35 ˚C until it reached a constant weight. Dry samples were finely grounded with high speed coffee grinder to 1-2 mm pieces. The fine, dried samples (1 g) were extracted with 20 ml solvent for solvent and ultrasound assisted extraction, 2 g dried samples were extracted with 40 ml solvent for microwave assisted extraction. Extractions were performed using Box-Benken design at 15 different experiment condition. Extracts centrifuged 20 min at 2500 rpm than the upper layer was taken and stored at -20˚C until they were analysed.  All samples were extracted in duplicate and average values were reported. Total phenolic content was determined with Folin-Ciocalteu method. The results were expressed as mg Gallic acid equivalent (GAE) per 100 g DW sample.  Total flavonoid analysis was done according to aluminum chloride colorimetric method. The results were expressed as milligrams of (+)-catechin equivalent (CE) per 100 g of DW sample. Total antioxidant capacity was analyzed with 2,2 diphenyl-1-picrylhydrazyl (DPPH)  analysis method. The results were expresed as  mg of Trolox equivalent (TE) per 100 g of DW of sample. Samples of each extraction were analyzed in triplicate and average values were reported. The optimal extraction conditions for solvent extraction method were 60% ethanol concentration, 53.93 ˚C extraction temperature, 84.84 min extraction time, while maximal theoretical total phenolic content was 3847.1 mg GAE/100 g DW, total flavonoid content was 3250.9 mg CE/100 g DW, total antioxidant capacity was 506.28 mg TEAC/100 g DW. The optimal extraction conditions for ultrasound assisted extraction method were 60% ethanol concentration, 60 ˚C extraction temperature, 38.03 min extraction time. Under these optimum conditions, predicted total phenolic content, total flavonoid content and DPPH values were as follows 4196 mg GAE/100 g DW, 3706 mg CE/100 g DW, 496.88 mg TEAC/100 g DW. The optimal extraction conditions for microwave assisted extraction method were 60% ethanol concentration, 518.18 Watt microwave power, 82.72 s extraction time, while maximal theoretical total phenolic content was 6271 mg GAE/100 g DW, total flavonoid content was 5033 mg CE/100 g DW, total antioxidant capacity was 493.22 mg TEAC/100 g DW. Total phenolic content and total flavonoid content of sage obtained by microwave assisted extraction at the optimum conditions were higher than those obtained by solvent extraction and ultrasound assisted extraction. Total antioxidant capacity with DPPH method of sage obtained by solvent extraction was higher than the other extraction techniques.
