Özet
Bu çalışmada, proteaz enzimi çitin ve çitosan üzerine 30oC’de kovalent bağlanma yöntemi ile immobilize edilmiştir. Çitin ve çitosan glutaraldehitle aktive edilerek çapraz bağlanma sağlanmıştır. Glutaraldehit miktarının ve tampon çözeltisinin (Tris-HCl) pH değerinin immobilizasyona etkisi incelenmiştir. Ayrıca, proteaz enzimi aljinat ve zeolit üzerine +4oC’de fiziksel adsorpsiyon yöntemi ile immobilize edilmiştir. En yüksek bağlanmış enzim miktarı ve immobilizasyon verimi kovalent bağlanma yönteminde çitin üzerine immobilizasyonda görülmüştür. İmmobilizasyon süresinin immobilizasyon verimi üzerinde direk bir etkisi olduğu belirlenmiştir. Serbest enzimle karşılaştırıldığında, immobilize enzimlerin toplam aktivitenin %80’ini tutabildiği belirlenmiştir. Çitin ve çitosan üzerine immobilize edilen proteaz enzimlerinin optimum sıcaklıkları 60oC olup serbest proteaz enziminden (50oC) daha yüksektir. Aljinat üzerine immobilize edilen proteaz enziminin optimum sıcaklığı ise 40oC olup serbest proteaz enziminden daha düşüktür. Çitosan, çitin, aljinat ve zeolit üzerine immobilize edilen enzimlerin optimum pH değerleri sırasıyla 9.5, 11.5, 10.0, 8.4’tür. İmmobilize proteaz enzimlerinin ticari bir deterjan ile %90 uyumluluk gösterdikleri belirlenmiştir. Çitosan, çitin, aljinat ve zeolit üzerine immobilize edilen enzimlerin 3 ay depolama sonrası orjinal aktivitelerinin %75, %72, %64 ve%59’unu koruduğu belirlenmiştir. Ayrıca, immobilizasyon prosesleri ile enzim sürekli sistemlere uygun hale getirilmiştir.
In this study, protease enzyme was immobilized on chitin and chitosan by covalent binding method at 30oC. Chitin and chitosan were activated with glutaraldehyde in order to obtain cross linking. The effect of the rate of glutaraldehyde and buffer pH (Tris-HCl) was investigated. Also, the protease enzyme was immobilized on alginate and zeolite by physical adsorption method at + 4oC. The highest bound enzyme and immobilization yield were found in the case of with chitin by covalent binding method. It was found that the immobilization time was directly affected to the immobilization yield. The immobilized enzyme retained 80% of total activity compared with the soluble enzyme. The optimum temperature of the resulting immobilized protease on chitin and chitosan was measured as 60oC being higher than that of free protease (50oC). The optimum temperature of the resulting immobilized protease on alginate was 40oC and it was lower than that of free protease. Optimum pH of the immobilized enzyme on chitosan, chitin, alginate and zeolite were 9.5, 11.5, 10.0, 8.4 respectively. Immobilized protease enzymes showed 90% compatibility with a commercial detergent. Proteases immobilized on chitosan, chitin, alginate and zeolite were maintained 75%, 72%, 64% and 59% of their original activities after 3 months storage. Also, the enzyme was made suitable for continuous systems by immobilization processes.
