Özet
Beş farklı tekstil boyasını gideren tek odalı hava-katot mikrobiyal yakıt hücrelerindeki (MYH) katot mikrobiyal film toplulukları 16S rRNA gene dayalı metotlarla tespit edilmiştir. Aynı aşı, elektrot materyalleri, MYH tasarımları ve çalıştırma koşullarına rağmen MYH’ler arası maksimum güçler farklılık göstermiştir. PZR-DGJE analizleri sonucunda reaktörlere tekstil boyalarının verilmesiyle mikrobiyal topluluklarda önemli miktarda değişimler görülmüştür. Oluşturulan klon kütüphanelerinin analizlerinde Alfa-, Beta- ve Gammaproteobakterilere ve Aktinobakterlere ait muhtelif bakteriyel gruplar tespit edilmiştir. Bu gruplar içerisinde Aquamicrobium, Mesorhizobium, Ochrobactrum, Thauera, Paracoccus, Achromabacter, Chelatacoccus ve Methanobacterium ilişkili türler klon frekanslarına gore ana türler olarak belirlenmiştir. Geçmiş çalışmalardan farklı olarak, elde edilen sonuçlar katot mikrobiyal film topluluklarının da tek odalı MYH’lerde güç üretiminde etkin olabileceğini göstermektedir. Rhizobiales klonlarının %59.5’ini oluşturan Mesorhizobium türü mikrobiyal filmde en sık rastlanan bakteridir. Bu durum, türün, aerobik solunumda oksijeni terminal elektron alıcısı olarak kullanabilmesi ile örtüşmektedir. Türlerin metabolik aktiviteleri ile MYH performansının ilişkilendirilmesi konusunda yapılacak çalışmalar daha verimli enerji üretilmesine olanak sağlayabilir.
Microbial communities on cathode biofilm of a single-chamber air-cathode microbial fuel cells decolorizing five different textile dyes were identified using 16s rRNA gene dependent methods. Despite the identical inocula, electrode materials, MFC architecture and operational conditions used, the maximum power varied among the devices. PCR-DGGE analysis demonstrated a major change in microbial communities upon introduction of textile dyes to the reactors. Clone library analysis showed the presence of diverse groups of Bacteria which belong to Alpha-, Beta-, Gamma-proteobacteria and Actinobacteria. Aquamicrobium, Mesorhizobium, Ochrobactrum, Thauera, Paracoccus, Achromabacter, Chelatacoccus and Methanobacterium affiliated phylotypes were the genera encountered most frequently. As opposed to the previous studies, results suggested that cathode biofilm in single-chamber MFCs may also have an active role in power generation. Mesorhizobium spp. comprised 59.5% of the Rhizobiales clones and was the most abundant bacteria. This agrees with their ability to use oxygen as their terminal electron acceptor during aerobic respiration. Further studies are needed for a better understanding of the relationship between metabolic activities of the individual species and the MFC performance in order to improve energy yields.
