Spg4 Ve Katnb1 Gen Anlatımlarının Elk1 Transkripsiyon Faktörü İle Düzenlenmesi

Özet
Hücre iskeletinin bir elemanı olan mikrotübüller, hücre içi moleküler taşınımı, mitoz sırasında kromozomların ayrılmas ve hücre şeklinin oluşturulması gibi birçok hücresel işlemde görev alır. Özellikle nöronların aksonal ve dendiritik uzantılarının oluşumunda temel mekanizma mikrotübüllerin kesimidir. Nöronlarda bu kesimi sağlayan spastin ve katanin adı verilen iki enzim bulunur. Spastin SPG4 geni tarafından kodlanırken, p60 ve p80 altbirimlerine sahip olan katanin sırasıyla KATNA1 ve KATNB1 genleri tarafından kodlanır. Enzimatik aktiviteye p60 katanin sahipken; p80-katanin, p60-kataninin aktivitesini düzenler. Bu proteinlerin miktarları gen anlatımlarının düzenlenmesiyle doğrudan ilişkilidir. Bir transkripsiyon faktörü olan Elk1 hem SPG4 hem de KATNB1 genlerinin anlatımlarının düzenlenmesinde görev alır. Bu çalışmada Elk1 transkripsiyon faktörlerinin SPG4 ve KATNB1 gen promotörleri üzerindeki etkisi araştırılmıştır. Elk1’in SPG4 promotörü üzerindeki baskılayıcı etkisi lüsiferaz bildirici gen testi ile belirlenmiştir. Bu baskılanmanın Elk1 ‘in SUMO modifikasyonundan dolayı olduğunu belirlemek için SUMO modifikasyon bölgeleri çıkarılmış yeni dizi oluşturulmuştur. Ayrıca PMA kimyasalı ile de SUMO modifikasyonu yok edilmiştir. Her iki durumda da baskılayıcı etkinin tersine çevrilmiştir ve lüsiferaz bildirici gen testi ile gösterilmiştir. Ayrıca sonuçlar gerçek zamanlı PZR ve immünblotlama teknikleriyle transkripsiyonel ve translasyonel düzeyde de doğrulanmıştır. Elk1, KATNB1 gen promotörü üzerindeki aktivatör etkisi lüsiferaz bildirici gen testi ile belirlenmiştir. Bu aktivatör etki KCl uygulaması ile yok edilmiş ve tüm deneyler gerçek zamanlı PZR yöntemi ile transkripsiyon, immunblotlama ve Western blotlama tekniğiyle translasyon seviyesinde doğrulanmıştır.
Microtubules are member of cytoskeleton and have roles in some cellular activities such as vesicular transport, chromosome segregation during mitosis and formation of cell shape. One of the major mechanisms that takes role in formation of axon and dendirites in neuron is microtubule severing. There are two microtubule severing enzymes called spastin and katanin. Spastin is encoded by SPG4 gene. Katanin is a heterodimer and its subunits p60 and p80 are encoded KATNA1 and KATNB1genes, respectively. p60-katanin has the enzymatic activity for severing microtubules, while p80-katanin regulates role of p60-katanin. The amounts of these proteins in the cell are related with regulation of gene expression. Elk1, a transcription factor, has role in regulation of both SPG4 and KATNB1 gene expressions. In this study effect of Elk1 on SPG4 and KATNB1 gene expressions was investigated. The respressor effect of Elk1 on SPG4 gene promoter was identified by using luciferase reporter assay. In order to determine this repression is due to SUMOylation of Elk1, sequence including SUMOylation site on Elk were deleted. On the other hand Elk1 was treated with PMA for deSUMOylation. In both cases repressor activity of Elk1 was reversed and demonstrated with using luciferase reporter assay. These results were confirmed by using real-time PCR and immunoblotting. The activator role of Elk1 on KATNB1 gene promoter was identified with using luciferase reporter assay. These acitvator effect was diminished with KCl treatment and these results were confirmed with real-time PCR, immunoblotting and Western blotting in transcriptional and translational levels.
