dc.description.abstractSaccharomyces cerevisiae’de etanol direnci, etanol üretimini etkileyen en önemli faktörlerden biri olduğu için, endüstriyel etanol üretim biyoprosesleri konusunda çalışan pek çok araştırma grubu için önemli bir konudur. Etanol direncinin moleküler mekanizması karmaşık olduğundan, bir veya birkaç gen üzerinde yapılacak değişikliklere dayalı rasyonel metabolizma mühendisliği yaklaşımı ile bu mekanizmanın aydınlatılması mümkün görünmemektedir. Bu çalışmada, etanole dirençli S. cerevisiae mutantlarının elde edilmesi ve karakterizasyonu için, bir tersine metabolizma mühendisliği stratejisi olan evrimsel mühendislik yöntemi benimsenmiştir. Yüksek etanol direncine sahip mutant bireyler, dereceli olarak artan etanol stres koşulları altında elde edilmiştir. Sonuçlar, benimsenen stratejinin istenilen özellikte kültürler elde etmede oldukça etkili bir yöntem olduğunu göstermiştir. Elde edilen dirençli mayaların karakterizasyonu da gerçekleştirilmiştir. Etanol stresi altında etanole dirençli mayalarda eşey tipi dönüşümü ve buna bağlı diploitleşmenin meydana gelişi, endüstriyel maya suşlarında olduğu gibi, etanole dirençli laboratuvar suşlarında da birden fazla kromozom kopyası bulundurma (multi-ploidy) eğilimini ortaya koymuştur. Bu çalışma, dereceli olarak artan etanol stresi koşullarına cevaben, S. cerevisiae hücrelerinde eşey tipi dönüşümünün tetiklendiğini ortaya koyan öncü bir çalışmadır.
dc.description.abstractAs ethanol resistance in Saccharomyces cerevisiae is one of the most important factors affecting ethanol production, it is an important issue for many research groups working on industrial ethanol production bioprocesses. Because of the complex molecular mechanism of ethanol resistance, it does not seem to be possible to clarify this mechanism by using rational metabolic engineering approach that is based on manipulations on one or a few genes. In this study, evolutionary engineering, an inverse metabolic engineering strategy, was adopted to obtain and characterize ethanol resistant S. cerevisiae mutants. Mutant individuals with high ethanol resistance were obtained under gradually increasing ethanol stress conditions. The results showed that the adopted strategy is an efficient method to obtain cultures with desired characteristics. The resistant yeasts obtained were also characterized. The emergence of mating type switching and diploidization in ethanol resistant yeasts under ethanol stress conditions pointed out the tendency of the ethanol-resistant laboratory strains, like industrial yeast strains, for multi-ploidy. This study constitutes a pioneering work implying the induction of mating type switching in response to gradually increasing levels of ethanol stress in S. cerevisiae cells.
