Tau hiperfosforilasyonu ve amiloid beta birikimleri nörodejenerasyonun ana belirteçleri olarak kabul edilmektedir. Bunlara ek olarak, anormal hücre döngüsüyle ilişkili olaylar nörodejeneratif patolojinin önemli göstergelerinden biri haline gelmektedir. Fosfo-Ser/Thr-Pro spesifik prolil-izomeraz, Pin1, nöronlarda anormal hücre döngüsüyle ilgili önemli bir hedef olarak belirlenmiştir. Pin1, hücre döngüsünün birçok noktasında görev almakta ve bu yüzden birçok nörodejeneratif hastalıkların patolojisi ile ilişkili bir noktada bulunmaktadır. Pin1’in oksidatif disregülasyonu hem normal tau fonksiyonlarının bozulmasına ve tau hiperfosforilasyonuna hem de hücre döngüsü kontrolünün yitirilmesine neden olmaktadır. Siklin D1, A, B1, PCNA ve Siklin Bağımlı Kinazlar gibi birçok hücre döngüsü ilişkili proteinin nörodejeneratif şartlarda merkezi sinir sisteminde varlığı gösterilmiştir. Daha da önemlisi, Pin1’in bu proteinlerin birçoğu ile fiziksel etkileşimi belirlenmiştir. Bu nedenle Pin1’in nöronlarda anormal hücre döngüsü aktivasyonu ile ilgisinin anlaşılabilmesi yeni terapötik yöntemler geliştirebilmek açısından önemlidir. Bu çalışmada, Pin1, kendi selektif inhibitörü olan juglon ile inhibe edilerek bunun hipokampal nöronlarda hücre döngüsü sürecine etkisi incelenmiştir. Pin1’in juglone inhibisyonu hipokampal nöronlarda aksonal retraksiyon, siklin D1 seviyesinde artış ve siklinlerin sub-selüler lokalizasyonlarında değişime neden olmuş ve Alzheimer Hastalığı benzeri bir patoloji oluşturmuştur. Bu nedenle, Pin1’in nöronlardaki fonksiyon kaybının, nörodejeneratif patolojinin oluşuma önemli katkıları olabilir.
Tau hyperphosphorylation and amyloid beta deposition are considered as the hallmarks of neurodegeneration. In addition to these, abnormal cell cycle events are also increasingly becoming important attributes of neurodegenerative pathology. The phospho-Ser/Thr-Pro specific prolyl-isomerase, Pin1 is becoming a crucial target in relation to its functions regarding these abnormal cell cycle events in neurons. Pin1 is majorly involved in many aspects of cell cycle regulation and holds the potential to be a crucial target provoking the pathology behind many neurodegenerative diseases. Oxidative dysregulation of Pin1 affects not only normal tau regulation, eventually causing tangle formation, but also cell cycle regulation in neurons. Presence of cell cycle proteins such as cyclin D1, cyclin A, cyclin B1, PCNA and Cyclin Dependent Kinases in many neurodegenerative diseases has been shown despite of the common notion regarding the absence of cell cycle related proteins in central nervous system. Importantly, many of these proteins have physical interactions with Pin1. Hence, understanding Pin1’s role in abnormal cell cycle re-entry is critical in terms of finding a new ways of approach for the future therapeutic options treating neurodegenerative pathologies. This study shows that inhibition of Pin1 by its selective inhibitor juglone, leads up-regulation of cyclin D1, axonal retraction and a change in sub-cellular localizations of cyclins in cultured rat hippocampal neurons also producing an Alzheimer’s Disease-like pathology. Therefore, Pin1 dysregulation may have profound effects in the progress of neurodegenerative pathology, making it crucial target behind neurodegenerative pathology.
