Özet
Bu çalışmada, E. coli ’ nin fizyolojik özellikleri ve biyokimyasal depolama mekanizmaları incelenmiştir. Değişik kültivasyon koşullarının büyüme ve depolama mekanizmaları üzerindeki etkisinin karşılaştırılabilmesi için respirometrik analizlerle birlikte kesikli, yarı kesikli ve ardışık kesikli reaktör sistemleri kullanılmıştır. Değişik karbon kaynaklarının büyüme ve depolama mekanizmaları üzerindeki etkisinin anlaşılabilmesi için çeşitli konsantrasyonlarda karbon kaynağı olarak glikoz veya gliserin içeren standart minimal (M9)besiyeri kullanılmıştır. Azot limitasyonunun büyüme ve depolama mekanizmaları üzerindeki etkisinin belirlenebilmesi için değişik derişimlerde NH4Cl içeren standart minimal besiyeri ile kesikli erlen deneyleri uygulanmıştır. Glikozun tek karbon kaynağı olarak kullanıldığı yarı kesikli ve ardışık kesikli kültivasyon deneyleri sırasında ve NH4Cl’nin tek azot kaynağı olarak kullanıldığı azot limitasyon deneylerinde glikojen depolama ve tüketimi gözlenmiştir. Diğer taraftan gliserinin tek karbon kaynağı olarak kullanıldığı kesikli erlen deneyleri sonucunda glikojen depolanmasına rastlanmamıştır. Glikozun ve gliserinin tek karbon kaynağı olarak kullanıldığı deneysel koşulların hiçbirinde PHB depolanması tespit edilmemiştir.
In this study, physiology and the biochemical storage characteristics of E. coli was investigated. Batch, fed-batch and sequencing batch cultivations were performed along with respirometric measurements to compare the effects of different cultivation conditions on growth and storage mechanisms. Various concentrations of either glucose or glycerol were used as the sole carbon source of the standard minimal medium (M9) to investigate the effects of different carbon sources on growth and storage. Batch cultivations with M9 without any nitrogen source and with varying amounts of NH4Cl as the sole nitrogen source were conducted to analyze the effects of nitrogen limitation on the storage and growth behavior of E. coli. During fed-batch and SBR cultivations of E. coli with glucose as the sole carbon source, glycogen storage and consumption was observed. Also at batch shake flask experiments where various NH4Cl concentrations were used for nitrogen starvation, glycogen production was observed. On the other hand, no glycogen formation was measured with various concentrations of glycerol in batch shake flask and bioreactor cultivations. No storage of PHB was observed under any of the cultivation conditions with neither glycerol nor glucose as the sole carbon source.
