Özet
Bu çalışmada, Türkiye’nin güneyinde Afyon, Denizli, Burdur il sınırları arasında yer alan aşırı tuzlu (200g/l) bir karakter gösteren Acıgöl’ün bakteriyel topluluğu belirlenmiş ve bu türlerin biyomineralizasyon üzerindeki etkileri belirlenmiştir. Acıgöl’den alınan toprak örnekleri kullanılarak %3 ve %6 NaCl içeren Nutrient Broth besiyerinde zenginleştirme kültürleri hazırlanmış ve bu kültürlerden hem tür analizleri hem de biyomineralizasyon deneyleri yapılmıştır. Göldeki bakteriyel çeşitliliğin ortaya koyulması için 16S rDNA PZR ( polimeraz zincir reaksiyonu ) metodu kullanılmıştır. Biyomineralizasyon deneyleri için gölün fizikokimyasal koşulları göz önünde bulundurularak özel bir besiyeri (%1 yeast extract, %0,1 glikoz, %0,5 pepton, % 4,5 NaCl ve 84 mM Mg+2, 11 mM Ca+2 g/L ) dizayn edilmiş ve deneyler dört farklı sodyum sülfat (Na2SO4) konsantrasyonu ( 0, 14, 28, 56 mM Na2SO4) olacak şekilde kurulmuştur. Hazırlanan besiyerleri zenginleştirme kültürleriyle aşılanmış ve 30°C’de 180 rpm’de çalkalamalı olarak inkübe edilmiştir. Oluşan mineraller saflaştırılarak XRD ve SEM ile karakterize edilmiştir. Deneyler sonucunda Acıgöl’de Halomonas, Idiomarine, Virgibacillus ve Uncultured bacteirum gibi halofil (tuz seven) türlerin baskın olduğu görülmüş ve bu türlerin Monohidrokalsit, Dypingite, Struvite, Hidromanyezit gibi minerallerin oluşumunda etkili olduğu kontrollü deneylerle gösterilmiştir.
In this study, Lake Acigol’s bacterial population was determined and these microorganisms influence on biomineralization is exhibited. Lake Acigol which is located between Afyon, Denizli and Burdur city boundaries and located in southern Turkey, is characterized by its high salinity concentration (200 g/l). Sediment samples were collected from Lake Acigol and enrichment cultures which were consist of 3% and 6% NaCl containing Nutrient Broth and sediment samples were carried out. Enrichment cultures were used for both determinations of bacterial population and biomimeralization experiments. Bacterial population was determined by 16S rDNA PCR (Polymerase Chain Reaction) method. A Special medium (%1 yeast extract, %0,1 glikoz, %0,5 pepton, % 4,5 NaCl ve 84 mM Mg+2, 11 mM Ca+2 g/L ) were designed for biomineralization experiments according to physicochemical conditions of Lake Acigol and experiments were carried out four different sodium sulfate ( Na2SO4) concentration (0, 14, 28, 56 mM Na2SO4). Prepared medium were inoculated with sediment samples and incubated at 30°C, with 180 rpm by shaking. Biominerals were purified and characterized by XRD and SEM. According to results, Halomonas, Idiomarine, Virgibacillus and Uncultured bacteirum are dominant in Lake Acigol and these microorganism were effective on producing biominerals such as Monohidrokalsit, Dypingite, Struvite, Hidromanyezit.
