dc.description.abstract | Bu çalışmada, boraj yağından çörekotu bitkisel lipazı katalizörlüğünde enzimatik hidroliz yoluyla sağlık ve gıda alanlarında kullanımı ve önemi giderek artan γ – linolenik asitçe zengin ürünler eldesi amaçlanmıştır. Hidroliz reaksiyonlarında kullanılan enzimin çörekotu tohumlarından ekstraksiyonunda içersinde % 1 triton bulunan distile suyun en uygun ekstraksiyon çözücüsü olduğu ve bu çözücü sistemiyle elde edilmiş lipaz ekstraktından enzimin çöktürülmesinde de % 35 amonyum sülfat doygunluk derecesinde çalışılmasının uygun olacağı belirlenmiştir. Proses parametrelerinin (hidroliz süresi ve enzim konsantrasyonu) hidroliz reaksiyonu yürüyüşüne olan etkisi incelenmiş, özellikle 163 – 490 U/g yağ enzim konsantrasyonlarında yürütülmüş hidroliz reaksiyonlarında, γ – linolenik asidin hidroliz ürünlerinden Digliserid (DG) ve Trigliserid (TG) fraksiyonlarında zenginleştiği saptanmıştır. En iyi sonuçlar 326 U/g yağ enzim miktarında elde edilmiş olup, 8 saat sonunda, orijinal boraj yağında % 21,9 oranında bulunan γ – linolenik asidin TG ve DG fraksiyonlarındaki yüzdesi sırasıyla % 29,6 ve % 41,8’ e yükselmiştir. | tr_TR |
dc.description.abstract | In this study, to obtain γ – linolenic acid rich products by enzymatic hydrolysis of borage oil was aimed since γ – linolenic acid has become one of the important polyunsaturated fatty acids in health and food industry. Distilled water with % 1 triton was found to be the most suitable solvent for extraction and optimum ammonium sulphate saturation level was established as % 35 for the precipitation of the extracted enzyme. The effects of process parameters (hydrolysis time and enzyme concentration) on the course of hydrolysis reaction and on the enrichment of γ – linolenic acid in the hydrolysis products were investigated. γ – linolenic acid was concentrated in the Diglyceride (DG) and Triglyceride (TG) fractions of the hydrolysis products, particularly at 163 – 490 U/g oil enzyme concentrations. Best results were obtained at 326 U/g oil enzyme concentration. After 8 h, the level of γ – linolenic acid was raised from 21.9 % in the original borage oil to 29.6 % and 41.8 % in the TG and DG fractions, respectively. | en_US |
