Özet
Obezite problemi bilincinin gelişmesi ile, dünyada düşük kalorili yağların ve yağ ikame maddelerinin pazarı hızlı bir büyüme göstermektedir. Günümüzde karbonhidrat ve protein bazlı yağ ikame maddeleri, bazı gıda maddelerinde kullanılmaktadır. Ancak bu ikame maddeleri ısıl işleme dayanıklı değillerdir. Bu yüzden, pişirme ve kızartma işlemleri gerektiren gıda maddelerinin üretiminde, ısıl dayanıklı ve doğal bir yağın özelliklerini gösteren lipid bazlı yağ ikame maddelerinin kullanımı tek seçenek olmaktadır. Düşük kalorili yağlar ve yağ ikame maddeleri, esas olarak lipidlerin yeniden yapılandırılmalarıyla üretilebilmektedir. Düşük kalorili yapılandırılmış lipidler, 1,3-pozisyonlarında orta zincirli yağ asitleri (C8-C12) ve 2-pozisyonunda uzun zincirli yağ asitleri (C16-C22) bulunan triaçilgliserollerdir. Bu çalışmada, tripalmitinden (TP) düşük kalorili yağların üretiminde kullanılabilecek, hızlı metabolize olan kaprik asit (C10:0) ile zenginleştirilmiş triaçilgliserollerin enzimatik olarak eldesi amaçlanmıştır. Bu amaçla, tripalmitinin kaprik asit (KA) ile Thermomyces lanuginosa lipazı (Lipozyme TL IM) varlığında asidoliz reaksiyonu sistematik olarak incelenmiştir. Tepki Yüzey Metodolojisi uygulanarak, reaksiyon parametrelerinin (substrat oranı, enzim miktarı ve sürenin) tripalmitine katılan kaprik asit yüzdesi üzerine olan etkileri araştırılmış ve reaksiyon koşulları optimize edilmiştir. Optimizasyon için kullanılan Tepki-Yüzey Metodolojisi yöntemine göre 50 °C’de yapılan seri deneyler sonucunda, tripalmitine en fazla %35,01 kaprik asit katılabileceği belirlenmiştir. Bu istatistiksel analiz yöntemine göre optimum reaksiyon koşulları ise 8,9 saat reaksiyon süresi, %11,6 enzim miktarı ve 1/6,5 TP:KA mol oranıdır. TP:KA mol oranı, reaksiyon süresi ve enzim miktarının kaprik asit yüzdesine etkisini göstermek için varyans analizinden yararlanarak 2. dereceden polinom olan bir model denklem oluşturulmuştur. Elde edilen model denklemin regresyon katsayısı (R2) 0,99 olarak bulunmuştur. Bu da model denklemin güvenilir olduğunu göstermektedir.
With increasing awareness of the risks associated with obesity, a market for reduced calorie fats and fat replacers has opened up. Carbohydrate and protein-based fat replacers are currently available, but cannot be exposed to high temperatures. Therefore, lipid-based fat substitutes are the only option for use in cooking and frying applications and for mimicking all the attributes of a natural fat. Low calorie fats and fat replacers can be produced by restructuring lipids. In general, these structured lipids contain medium chain fatty acids (C8-C12) in the 1,3-positions and long chain fatty acids (C16-C22) in the 2-position of triacylglycerols. The aim of this project is the production of triacylglycerols enriched with capric acid (CA) to be used in the formulation of low calorie fats. For this purpose, the acidolysis reaction of tripalmitin (TP) with capric acid in the presence of Thermomyces lanuginosa lipase (Lipozyme TL IM) was studied in detail. The effects of reaction parameters (substrate ratio, amount of enzyme amd time) on the percentage of incorporated capric acid into tripalmitin were investigated and reaction conditions were optimized using Response Surface Methodology. Maximal incorporation of 35,01% capric acid into tripalmitin was predicted by Responce Surface Methodology when a substrate molar ratio of 1:6,5 of TP/CA, 11,6% enzyme amount and incubation time of 8,9 h were used. A second-order model was obtained to predict conversions as a function of time, enzyme percent and TP:CA mol ratio. The analysis of variance indicated that the second-order model was highly significant and adequate to represent the actual relationship between the response (CA content, %) and the significant variables with a satisfactory regression coefficient (R2) found as 0,99.
