BİTKİSEL KAYNAKLI ANTİMİKROBİYALLERİN GIDA KAYNAKLI BAZI PATOJEN MİKROORGANİZMALAR ÜZERİNDE ETKİLERİ Elvan ŞAHİN Doğal ve katkı maddesi içermeyen ürünlere yönelik artan tüketici talepleri, yeni üretim teknikleri ve doğal antimikrobiyallere olan ilgiyi arttırmıştır. Bu çalışmada bitkisel kaynakların gıdalarda mikrobiyal gelişmenin kontrol altına alınmasında kullanımını araştırmak üzere kimyon, maydonoz esansiyel yağları, paprika, zencefil, kimyon ve maydonoz oleoresinleri ve greyfurt çekirdeği ekstraktının (GSE) Bacillus cereus, E. coli O157:H7, Salmonella enteritidis gibi gıda kaynaklı bazı patojen bakteriler üzerinde inhibitör etkileri incelenmiştir. Araştırılan bitkisel kaynaklar içerisinde oleoresinler kağıt disk metodunda inhibitör etki göstermezken, maydonoz, kimyon esansiyel yağları ve GSE patojen üç mikroorganizmanın gelişimini olumsuz etkilemiştir. Agar dilüsyon yönteminde en düşük MIC değerleri GSE için saptanmış, bunu kimyon ve maydanoz esansiyel yağı izlemiştir. Ortamın yağ içeriğinin GSE’nin antimikrobiyal aktivitesi üzerindeki etkisini belirlemek üzere yapılan in vitro çalışmada emülsiyon oluşumu için kullanılan Tween 20’nin inhibitör etkiyi azalttığı, GSE ve yağın mikrobiyal populasyon üzerinde etkili olduğu saptanmıştır. In vitro çalışmalarda yüksek antimikrobiyal etki göstermesine rağmen GSE gıda matriksinde inhibitör etki gösterememiştir.
INHIBITORY EFFECTS OF PLANT DERIVED ANTIMICROBIALS ON SOME FOOD-BORNE PATHOGENIC BACTERIA Elvan ŞAHİN Increasing demand of food which is natural and free from additives, resulted in growing interest of natural antimicrobials. In this research, potential of essential oils of cumin and parsley, oleoresins of paprika, ginger, cumin and parsley, also grapefruit seed extract in controlling microbial growth was investigated on some food borne pathogenic bacteria such as Bacillus cereus, E. coli O157:H7, Salmonella enteritidis. Oleoresins did not inhibit pathogens in disk diffusion method, while grapefruit seed extract, cumin and parsley essential oils did. Having the lowest minimum inhibition concentration (MIC) GSE was followed by cumin and parsley essential oil. Inhibitor effect of GSE was investigated in the presence of oil in growth medium. It was found that using Tween 20, which was used for oil-in-water emulsion stability, decreased or destroyed inhibitor effect of GSE. Oil concentration was found statistically important and decreased antimicrobial effect of GSE. Although GSE was effective according to results that obtained in vitro, no antimicrobial effect was observed in food matrix.
