Abstract

Kobalt canlılar için gerekli bir metaldir ancak organizmaların ihtiyacından fazla alınması durumunda hücresel faaliyetlere zarar vermesi nedeniyle toksik etki yaratmaktadır. Kobaltın hücreye alımı, hücre içerisinde kullanılması ve hücrenin bu metale karşı gösterdiği direnç mekanizmaları diğer bir çok elzem metala oranla daha az bilinmektedir. Saccharomyces cerevisiae, moleküler biyoloji çalışmalarında ileri ökaryotlarla çok yakın ve benzer genetik yapısı nedeniyle tercih edilen bir mayadır. Bu tez çalışmasında kobalt direncine sahip S. cerevisiae mutant mayalarının eldesi, bu elementin biyolojik sistemlerde ve metabolizmada etkilerinin moleküler düzeyde çalışılabilmesi için gerçekleştirilmiş. Bu amaçla da, tersine metabolizma mühendisliğinin bir dalı olan evrimsel mühendislik yöntemi kullanılmıştır. Yapılan genetik ve transkriptomik analizler ardından kobalt direncinin başka metallerle de çok ilgili olabileceği gösterilmiş, özellikle demir homeostazisi ile kobalt direnci arasındaki yakın ilişki belirtilmiştir. Evrimsel mühendislik ile kobalta dirençli olan mutantların, metal ve diğer yolizlerinin hücre içerisinde birbirleri ile etkileşimlerini ortaya çıkarması açısından önemli bir çalışma olmuştur.
Cobalt is an essential transition metal however it can be highly toxic at elevated concentration to the living cells. Cobalt uptake into the cell, its usage inside the cytosol and the defence mechanisms of the organisms against cobalt is not known as much as other essential metals. Saccharomyces cerevisiae is a yeast species and its is widely used in molecular biology studies because of its conserved genetics to the higher eucaryotes. In this thesis, cobalt-resistant S.cerevisiae mutants were obtained in order to understand the role and effects of cobalt to the biological systems and metabolism. To this end, evolutionary engineering, which is a method of inverse metabolic engineering was accomplished. The genetic and transcriptomic analysis revealed that cobalt resistance might be related at high extent to other metals and especially iron homeostasis and cobalt resistance was found to be interrelated. This study could be considered as important in clarifying that metal resistance and other metabolic pathways in an organisms could have reciprocal influences.
