Abstract
Bu çalışmada, endüstride geniş uygulama alanı olan proteazın immobilizasyonu incelenmiştir. Bu amaçla ticari proteaz enzimi fiziksel adsorpsiyon, kovalent ve iyonik bağlanma yöntemleri kullanılarak farklı taşıyıcılar üzerine immobilize edilmiştir ve kovalent bağlanma metodu ile elde edilen immobilize enzimin karakterizasyonu yapılmıştır. Fiziksel adsorpsiyon metoduyla çitin ve tanin-çitosan üzerine immobilize edilmiş proteaz enziminin immobilizasyon verimleri sırasıyla, %44.2 ve %77.8 olarak bulunmuştur. Proteaz enziminin çitin, çitosan ve vermikulit üzerine kovalent bağlanma metoduyla immobilizasyonu sonucunda immobilizasyon verimleri sırasıyla, %84.3, %81.2 ve %54.8 olarak belirlenmiştir. İyonik bağlanma yönteminde ise en yüksek immobilizasyon verimi Ecteola23 Cellulose anyon değiştiricisi ile %26.5 olarak tespit edilmiştir. Çitin, çitosan ve vermikulit üzerine kovalent bağlanmayla immobilize edilmiş proteaz enzimi, serbest enzimin optimum sıcaklığının ve optimum pH’ının artmasını sağlamıştır. İmmobilize enzimlerin sıcaklık ve pH kararlılıklarında iyileşme tespit edilmiş, raf ömrü ve tekrarlı kullanılabilme deneyleri sonucunda kararlı davrandıkları belirlenmiştir. Ayrıca immobilize enzimlerin tümünün ticari deterjanlarla uyumlu oldukları gözlenmiştir.
In this study, the immobilization of protease, used in multiple areas in industry, was investigated. For this goal, commercial protease enzyme was immobilized on various carriers by the methods of physical adsorption, covalent and ionic binding and the characterization of the immobilized enzyme, which obtained by covalent binding, was analyzed. The immobilization yields of the proteases immobilized on chitin and tannin-chitosan by physical adsorption method were 44.2% and 77.8%, respectively. Covalent binding method by using chitin, chitosan and vermiculite resulted with the immobilization yields of 84.3%, 81.2% and 54.8%, respectively. The highest immobilization yield by using ionic binding method was achieved as 26.5% with the anion exchanger of Ecteola23 Cellulose. The covalently immobilized enzymes on chitin, chitosan and vermiculite increased the optimum temperature and optimum pH of the free enzyme. By the immobilization of protease on chitin, chitosan and vermiculite temperature stability and pH stability were improved. Covalently immobilized enzymes exhibited good storage stability and durability for repeated use. Furthermore, all the enzymes immobilized on chitin, chitosan and vermiculite exhibited good detergent stability with the commercial detergents.
