Abstract
Bu tez çalışmasında, nitrit ve nitrat anyonlarının kalitatif ve kantitatif tayini için kapiler elektroforez-örnek sıkıştırma yöntemi geliştirilmiştir. Asidik bir ayırma ortamı seçilerek kapiler içerisindeki güçlü elektroosmotik akışın hızı düşürülmüş ve elektroforetik mobiliteleri çok yüksek olan bu anyonların kısa bir sürede dedeksiyonu sağlanmıştır. Analiz yöntemi, kapiler içinde bir elektrik alan farkı yaratarak ve normal kapiler elektroforez injeksiyon hacmine göre daha yüksek bir hacimde injeksiyon yaparak, kapiler üzerinde örneğin derişiklendirilmesine dayanmaktadır. Bu yolla düşük hacimdeki injeksiyona oranla yöntemin hassasiyeti her iki anyon için de yaklaşık 30 kat arttırılmıştır. Yöntem, konserve ton, konserve uskumru ve sardalya örneklerinde nitrit ve nitrat anyonlarının tayininde başarıyla uygulanmıştır.
A capillary electrophoresis-sample stacking method was developed for the simultaneous analysis of nitrite and nitrate ions in this work. An acidic buffer was selected and electroosmotic flow was reduced with this way. Consequently, the anions were detected in a very short time. The sensitivity of the method was increased by applying a sample-stacking technique. When detection limits of both ions are compared between small volume injection and sample stacking modes, it was observed that the stacking mode resulted in a thirtyfold improvement in detection sensitivity for both anions. The method was successfully applied to canned fish samples, namely tuna, mackerel and sardine.
