Aspergillus İzolatlarının Tanısı, Mikotoksijenik Özelliklerinin Belirlenmesi, Sıcaklık Ve Sürenin İncirde Aflatoksin Ve Siklopiazonik Asit Oluşumuna Etkisi

Özet
Bu çalışmada, kurutma döneminde toplanan kuru incir örneklerinden izole edilmiş Aspergillus section Flavi üyelerinin PZR kullanılarak moleküler yöntemle identifikasyonu yapılmış ve izolatların in vitro koşullarda aflatoksin (AF), siklopiazonik asit (CPA) ve β-nitropropionik asit (BNP) oluşturma özellikleri belirlenmiştir. İkinci aşamada ise besiyerinde toksin üretme özelliklerine göre seçilen 5 adet izolat incire aşılanarak aflatoksin ve siklopiazonik asit üretimlerine sıcaklık ve sürenin etkisinin belirlenmesi amaçlanmıştır. Kuru incirden izole edilmiş 57 adet küf izolatının moleküler yöntemle tanımlanması ITS 1-5.8S-ITS 2 bölgesine spesifik primerler dizayn edilerek Polimeraz zincir reaksiyonu (PZR) ile yapılmıştır. Türlerin identifikasyonu 18S rDNA bölgesine ait sekans sonuçlarının BLAST’da bilinen ribozamal sekanslar ile karşılaştırılmasıyla gerçekleştirilmiştir. Küf izolatlarının 51’i Aspergillus flavus, 5’i Aspergillus parasiticus ve 1’i ise Aspergillus tamarii olarak tanımlanmıştır. İdentifikasyonda kullanılan ITS bölgesinin hedef alındığı moleküler yöntem 3 türü birbirinden ayırmada başarılı olmuştur. 57 adet izolatın AF, CPA ve BNP oluşturma özellikleri in vitro olarak besiyerinde incelenmiştir. İzolatların büyük oranda ve yüksek toksijenik özellikte olduğu belirlenmiştir. İncir aşılama çalışmalarında kullanılmak üzere in vitro AF ve CPA oluşturma özelliklerine göre 2 adet A. flavus, 2 adet A. parasiticus ve 1 adet A. tamarii izolatı seçilmiştir. İncirde AF ve CPA üretimine sıcaklık ve sürenin etkisinin istatistikî olarak çok önemli olduğu belirlenmiştir.
In this study, identification of Aspergillus section Flavi members by using PCR isolated from dried figs and determination of in vitro Aflatoxin (AF), cyclopiazonic acid (CPA) and β-nitropropionic acid (BNP) production capacity of the isolates were aimed. Regarding mycotoxigenic properties 5 different isolates were selected for studying effects of temperature and time on the aflatoxin and cyclopiazonic acid formation in figs. Molecular identification of 57 isolates from dried figs was conducted with designing specific primers for the ITS 1-5.8S-ITS 2 rDNA region using PCR. Identification of species was determined by comparing the result of sequences which belongs to partial 18S rDNA of the sequences with known ribosomal sequences using Basic Local Alignment Search Tool (BLAST). Out of a total 57 fungal isolates 5 of them identified as A. parasiticus, 1 of them identified as A. tamarii and 51 of them identified as A. flavus. ITS 1-ITS 2 target region for molecular identification is suitable for discrimination of three species. In vitro AF, CPA and BNP production properties of 57 isolates were studied on media and high percentages of the isolates were determined highly toxigenic. For the fig inoculation studies 2 A. flavus, 2 A. parasiticus and 1 A. tamarii isolates were selected according to in vitro AF and CPA production properties. Regarding temperature and time effects on both aflatoxin and cyclopiazonic acid production of the isolates in figs were significantly important.
