Özet
Bu çalışmada, kanola yağının kudret narı tohumu yağından elde edilen konjuge linolenik asit ile zenginleştirilmesi ve çeşitli fonksiyonel özelliklere sahip yapılandırılmış yağların üretimi amaçlanmıştır. Bu amaçla Thermomyces lanuginosa L. lipazı katalizörlüğünde enzimatik asidoliz reaksiyonları gerçekleştirilmiştir. Farklı reaksiyon koşullarının konjuge linolenik asit katılımı üzerine etkileri incelenmiş ve Tepki Yüzey Metodolojisi uygulanarak bu koşullar optimize edilmiştir. İki değişkenli ve üç seviyeli Yüzey Merkezli Küp Deney Tasarımı kullanılarak yürütülen deneyler sonucunda, optimum reaksiyon koşulları %10 enzim miktarı, 6,8:1 (konjuge linolenik asit:kanola yağı) substrat mol oranı, 54,2ºC reaksiyon sıcaklığı ve 3 saat reaksiyon süresi olarak saptanmıştır. Bu koşullarda, kanola yağından içeriğinde ağırlıkça %36,9 oranında konjuge linolenik asit bulunan sağlığa faydalı, fonksiyonel özellik taşıyan yapılandırılmış yağlar elde edilmiştir.
The main goal of this study is to produce structured lipids by the enzymatic acidolysis of canola oil and conjugated linolenic acid obtained from bitter gourd fatty acid blends. For this purpose, the interesterification reactions of canola oil with conjugated linolenic acid are carried out in the presence of an immobilized sn-1,3 specific lipase from Thermomyces lanuginosa L. The effects of different reaction conditions are evaluated and optimized by Response Surface Methodology. Two-factor, third-order Face Centered Cube Experimental Design is performed and the optimum reaction conditions are determined as 6,8:1 (conjugated linolenic acid:canola oil) substrate-molar ratio, 54,2ºC temperature, %10 enzyme load ve 3 hour reaction time. Under these conditions, the incorporation ratio of conjugated linolenic acid into canola oil is established as %36,9 and so healthier and more functional structured lipids are produced.
